چکیده

In the gill of the blue mussel Mytilus edulis, the lateral cilia, which are responsible for producing feeding currents, are neuronally activated (Aiello, 1960). The neurotransmitter is serotonin (Aiello, 1962), known to raise cAMP levels in the gill (Malanga and Poll, 1979). The functional target of cAMP action appears to be certain low molecular mass polypeptides that co-extract with the ciliary ATPase dynein (Stommel and Stephens, 1985). Phosphorylation of these polypeptides by the catalytic subunit of cAMP-dependent protein kinase is correlated with the acquisition of motility in ATP-reactivated cell models (Stephens and Stommel, 1989). Mounting evidence points to these target polypeptides as being dynein light chains and, even though the labile nature of Mytilus ciliary dynein has left this conclusion somewhat equivocal (Stephens and Prior, 1990a, 1991c), preliminary evidence for Spisula ciliary dynein strongly supports it (Stephens and Prior, 1991b). Serotonin also has been implicated in both natural spawning and sperm motility of bivalve molluscs (Lubet, 1981; Gibbons and Castagna, 1984; Hirai et al., 1988; Kadam and Koide, 1990), and there is now substantial pharmacological evidence for the presence of serotonin receptors on sperm of the surf clam Spisula solidissima (Kadam and Koide, 1990; Bandivdekar et al., 1992). The target of serotonin action, if any, at the level of the motile machinery of the sperm has not been studied. Since Mytilus and Spisula have both gill cilia and sperm flagella that can be activated in situ by serotonin, we carried out a comparative study of ciliary and flagellar dynein from these species, with emphasis on the target polypeptides of cAMP-dependent phos999 Journal of Cell Science 103, 999-1012 (1992) Printed in Great Britain © The Company of Biologists Limited 1992

THE EFFECT OF THEOPHYLLINE ON THE KINETICS OF cAMP-DEPENDENT PROTEIN KINASE CATALYTIC SUBUNIT, cAMP, PROTEIN KINASE INHIBITOR AND THEIR RELATIONSHIP IN LUNG TISSUE

We have investigated the effect of theophylline on the kinetics of the catalytic subunit of protein kinase and related factors in lung tissue. The results show that the point of highest concentration of the C subunit of protein kinase which is active in casein phosphorylation is at 3h of incubation time, but in the presence of 100 Ilg/ InL and 10µg/mL theophylline, this is shifted to I.S an...

Activation of Ciona sperm motility: phosphorylation of dynein polypeptides and effects of a tyrosine kinase inhibitor.

A high molecular mass dynein ATPase polypeptide and a 18-20 kDa dynein light chain of Ciona sperm flagella are phosphorylated during in vivo activation of motility or in vitro activation of motility by incubation with cyclic AMP. A similar level of phosphorylation of these proteins is obtained by incubation of washed, demembranated spermatozoa with catalytic subunit of cyclic AMP-dependent prot...

Study of PKA binding sites in cAMP-signaling pathway using structural protein-protein interaction networks

Backgroud: Protein-protein interaction, plays a key role in signal transduction in signaling pathways. Different approaches are used for prediction of these interactions including experimental and computational approaches. In conventional node-edge protein-protein interaction networks, we can only see which proteins interact but ‘structural networks’ show us how these proteins inter...

Elevation of cAMP Levels Inhibits Doxorubicin-Induced Apoptosis in Pre- B ALL NALM- 6 Cells Through Induction of BAD Phosphorylation and Inhibition of P53 Accumulation

Recognition of the molecular mechanisms of cAMP action against DNA damage-induced apoptosis can be useful to improve the efficacy of DNA damaging therapeutic agents. Considering the critical role of bcl-2-associated death promoter (BAD) and p53 proteins in DNA damage -induced apoptosis, the aim of this study was to assess the effect of cAMP-elevating agents on these proteins in doxorubicin-trea...
